Abstract:[Abstract: ObjectiveTo evaluate the killing effect of three kinds of lymphocytes (NK,NKT and γδT ) on nasopharyngeal carcinoma CNE1 cell in vitro for the potential clinical application. MethodsPeripheral blood mononuclear cells (PBMC) were drew from patients with nasopharyngeal carcinoma.The induced NK,NKT and γδT lymphocytes from PBMC were cultured for 14 days and cell phenotype was identified with flow cytometry method.CNE1 cell of nasopharyngeal carcinoma served as the target,three kinds of phenotypeconfirmed lymphocytes on the CNE1 respectively.The killing effects of NK,NKT and γδT lymphocytes were evaluated by real time cellular analysis (RTCA) method.The killing rates of the three kinds of lymphocytes on target cell were compared. ResultsThe three kinds of phenotypeconfirmed lymphocytes from the patients were obtained successfully.All three kinds of lymphocytes with various effect/target ratios can cause rapid cellular apoptosis of the target CNE1 cell in vitro tested by RTCA method.The killing rates of NKT lymphocyte on target CNE1 cell at 1,4 and 8 hours in vitro were significantly stronger than those of NK和γδT lymphocytes. ConclusionThe NKT and γδT lymphocytes have killing effect on nasopharyngeal carcinoma cell in vitro,the NKT lymphocyte is of mostsignificant effect and suggested that the three kinds of lymphocytes have the value of potential clinical application on biotherapy of nasopharyngeal carcinoma in the future.
