Detection on the content of heptaene menadione in health food by high-performance liquid chromatography method

Abstract:Objective To establish a method for detection the content of heptaene menadione (MK-7) of vitamin K2 in health food by high-performance liquid chromatography method. Methods The samples were extracted with isopropanol and analyzed by high-performance liquid chromatography, separated by C18 column, using methanol + isopropanol + 0.1% acetic acid water = 485 + 500 + 15(methanol,isopropanol and 0.1% acetic acid water was 485,500 and 15)as mobile phase, detected by PDA detector and quantified by external standard method. Results The detection limit was 0.02 mg/kg of the method, the linear ranges were 0.50 mg/L - 10.00 mg/L, the correlation coefficient was 0.9997, adding standard recovery rate was 95.0% - 98.3%, and the relative standard deviation (RSD) was 2.4% in the repeatability test. Conclusion This method is simple, accurate and reliable, and suitable for detecting the content of MK-7 in vitamin K2.
